ABSTRACT Objective: To determine the concentration of alpha-tocopherol in umbilical cord serum of full-term and preterm newborns, in order to assess the nutritional status of both groups in relation to the vitamin and its possible correlation with intrauterine growth. Methods: A cross-sectional observational study conducted with 140 newborns, of which 64 were preterm and 76 were full-term. They did not have any malformations, they came from healthy mothers, who were nonsmokers, and delivered a single baby. Intrauterine growth was evaluated by weight-to-gestational age at birth, using Intergrowth-21st. Thealpha-tocopherol levels of umbilical cord serum were analyzed by High Performance Liquid Chromatography. Results: The mean concentration of alpha-tocopherol in umbilical cord serum for preterm and full-term infants was 263.3±129.5 and 247.0±147.6 µg/dL (p=0.494). In the preterm group, 23% were small for gestational age, whereas in the full-term group, this percentage was only 7% (p=0.017). Low levels of vitamin E were found in 95.3% of preterm infants and 92.1% of full-term infants. There was no correlation between alpha-tocopherol levels and weight to gestational age Z score (p=0.951). Conclusions: No association was found between alpha-tocopherol levels and weight to gestational age at birth. Intrauterine growth restriction was more frequent in preterm infants and most infants had low levels of vitamin E at the time of delivery.


RESUMO Objetivo: Determinar a concentração de alfatocoferol em soro de cordão umbilical de recém-nascidos a termo e pré-termo, a fim de avaliar o estado nutricional de ambos os grupos com relação a essa vitamina e sua possível correlação sobre o crescimento intrauterino. Métodos: Estudo observacional de caráter transversal realizado com 140 recém-nascidos, 64 pré-termo e 76 a termo, sem malformações, oriundos de mães saudáveis, não fumantes e com parto de concepto único. O crescimento intrauterino foi avaliado pelo índice peso por idade gestacional ao nascer, utilizando a Intergrowth-21st. Os níveis de alfatocoferol do soro do cordão umbilical foram analisados por cromatografia líquida de alta eficiência. Resultados: A concentração média de alfatocoferol no soro do cordão umbilical para recém-nascidos pré-termo e a termo foi de, respectivamente, 263,3±129,5 e 247,0±147,6 µg/dL (p=0,494). Baixos níveis de vitamina E foram encontrados em 95,3% dos prematuros e em 92,1% dos neonatos a termo. Nogrupo pré-termo, 23% eram pequenos para a idade gestacional, enquanto no grupo a termo esse percentual foi de apenas 7% (p=0,017). Não houve correlação entre os níveis de alfatocoferol e o escore Z de peso para idade gestacional (p=0,951). Conclusões: Não foi encontrada associação entre os níveis de alfatocoferol e a adequação do peso à idade gestacional ao nascer. A restrição do crescimento intrauterino foi mais frequente nos nascidos pré-termo, e a maioria dos recém-nascidos apresentou níveis baixos de vitamina E no momento do parto.

Abstract Objective: The purpose of this study was to evaluate the antioxidant status of plasma vitamin E and plasma and intracellular coenzyme Q10 in children with type 1 diabetes. Method: This case-control study was conducted on 72 children with type 1 diabetes and compared to 48 healthy children, who were age, sex, and ethnicity-matched. The diabetic children were divided according to their glycosylated hemoglobin (A1c %) into two groups: poor and good glycemic control groups. All children underwent full history taking, clinical examination, and laboratory measurement of complete blood count, A1c %, plasma cholesterol, triglycerides, and vitamin E levels and coenzyme Q10 levels in plasma, erythrocytes, and platelets. Results: Children with poor glycemic control showed significantly higher plasma vitamin E, coenzyme Q10, triglycerides, low-density lipoproteins, waist circumference/height ratio, cholesterol levels, and lower high-density lipoproteins and platelet coenzyme Q10 redox status in comparison to those with good glycemic control and the control group (p < 0.05). Plasma coenzyme Q10 showed a positive correlation with the duration of type 1 diabetes, triglycerides, cholesterol, vitamin E, and A1c %, and negative correlation with the age of the diabetic group (p < 0.05). The platelet redox status showed a negative correlation with the A1c % levels (r = −0.31; p = 0.022) and the duration of type 1 diabetes (r = −0.35, p = 0.012). Conclusion: Patients with type 1 diabetes, especially poorly controlled, had elevation of plasma vitamin E and coenzyme Q10 levels and decreased platelet redox status of coenzyme Q10, which may be an indicator of increased oxidative stress.


Resumo Objetivo: Avaliar o estado antioxidante da vitamina E no plasma e da coenzima Q10 no plasma e intracelular em crianças com diabetes tipo 1. Método: Este estudo caso-controle realizado em com 72 crianças com diabetes tipo 1 comparadas por idade, sexo e etnia de 58 crianças saudáveis. As crianças diabéticas foram divididas em dois grupos de acordo com sua hemoglobina glicosilada (A1c %): grupos de controle glicêmico bom e baixo. Todas as crianças foram submetidas a anamnese total, exame clínico e laboratorial para hemograma completo, A1c %, colesterol no plasma, triglicerídeos e níveis de vitamina E e níveis de coenzima Q10 no plasma, eritrócitos e plaquetas. Resultados: As crianças com baixo controle glicêmico mostraram nível de vitamina E no plasma significativamente maior, coenzima Q10, triglicerídeos, lipoproteína de baixa densidade, proporção da circunferência da cintura/estatura e níveis de colesterol e menor nível de lipoproteína de alta densidade e estado redox da coenzima Q10 em comparação aos com bom controle glicêmico e com o grupo de controle (p < 0,05). A coenzima Q10 no plasma mostrou correlação positiva com a duração da diabetes tipo 1, triglicerídeos, colesterol, vitamina E e A1c % e correlação negativa com a idade do grupo diabético (p < 0,05). O estado redox das plaquetas mostrou correlação negativa com os níveis de A1c % (r = -0,31; p = 0,022) e a duração da diabetes tipo 1 (r = -0,35, p = 0,012). Conclusão: Os pacientes com diabetes tipo 1, principalmente mal controlados, apresentaram aumento nos níveis de vitamina E no plasma e coenzima Q10 e redução no estado redox das plaquetas da coenzima Q10 que podem indicar aumento do estresse oxidativo.

Globally, about 3.5 million women are living with genitourinary fistula, a miserable condition. Obstetric Vesicovaginal fistula (VVF) is exceptionally rare in developed part of the world, where it is mostly due to gynaecological cause. The study was conducted to provide baseline data on vitamin A and vitamin E levels among Vesico - Vaginal Fistula (VVF) patients and to establish their socio-demographic characteristics. Some selected vitamin levels in patients with Vesico - Vaginal Fistula (VVF) in Jos, Northern Nigeria were investigated using spectrophotometric method. Samples were collected from VVF patients undergoing treatments at the Evangel Hospital, Jos and the controls were non pregnant but parous women of similar social status who were willing to participate in the study. A total of 97 participants made up of 67 VVF patients and 30 controls were involved in the study. The results revealed that only 1 clients (1.49%) was mulliparous, 50 (74.64%) and 16 (23.88%) clients were multiparous and grand multiparous respectively out of 67 (100%) clients. The results of social class distribution in both clients and control is in the order of 5>4>3, 2>1. The levels of vitamin A in VVF clients showed no significant change (p>0.05) when compared to control while that of vitamin E showed significant change (p<0.05). The results of viatamin A and E in respect to age, social class and parity showed significant difference (p<0.05) in the VVF patients than the control except for those with age 25 - 29 and 40 - 44 years as well as social class 1 and 5. The study therefore highlights need for determination of reference values of vitamins levels among African population


A nivel mundial, alrededor de 3,5 millones de mujeres viven con fístula genitourinaria. La fístula vesicovaginal (FVV) obstétrica es excepcionalmente rara en el mundo desarrollado, donde se debe principalmente a causas ginecológica. Este estudio se realizó para proporcionar datos de referencia sobre los niveles de vitamina A y vitamina E entre los pacientes con FVV y para establecer sus características sociodemográficas. Los niveles de vitamina A y E en pacientes con VVF en Jos, norte de Nigeria se investigaron mediante el método espectrofotométrico. Se recogieron muestras de pacientes con FVV que se sometieron a tratamientos en el Evangel Hospital, Jos y un grupo control de paciente, no embarazadas del mismo estatus socioeconómico que estaban dispuestas a participar en el estudio. Un total de 97 participantes formados por 67 pacientes con FVV y 30 controles participaron en el estudio. Los resultados revelaron que solo 1 paciente (1.49%) era nulípara, 50 (74.64%) y 16 (23.88%) de las pacientes eran multíparas y multíparas grandes, respectivamente. Los resultados de la distribución socioeconómica en el grupo de embarazadas y el control fue en orden de 5> 4> 3, 2> 1. Los niveles de vitamina A en paciente con FVV no mostraron diferencias significativas (p>0.05) en comparación con el control, mientras que los niveles de vitamina E sí mostraron diferencias significativas (p<0.05). Los resultados de la vitamina A y E con respecto a la edad, clase socioeconómica y paridad mostraron una diferencia significativa (p<0.05) en los pacientes con FVV respecto control, excepto para los de 25-29 y 40-44 años, así como para la clase social 1 y 5. Por lo tanto, el estudio destaca la necesidad de determinar los valores de referencia de los niveles de vitaminas en la población africana.

Objective Bone loss has been established as a major extra-intestinal complication of short bowel syndrome (SBS). The purpose of this study was to correlate bone mineral density (BMD) with body mass index (BMI), serum vitamin and mineral levels in patients with SBS.Material and methods The study was conducted on 13 patients (8 male and 5 female, 54.7 ± 11.4 years) with SBS (residual small bowel length of 10 to 100 cm). We determined the food ingestion, anthropometry, serum levels of vitamins C, A, D, E and K, as well as serum and urinary levels of phosphorus and calcium. BMD was measured by dual-energy x-ray absorptiometry (DXA).Results Osteopenia and osteoporosis was diagnosed in all but one SBS patient. Serum levels of vitamin D were low in all volunteers. Sixty-one percent of patients had vitamin E deficiency; hypovitaminosis A and C occurred in one subject. BMI and C, E and K vitamin serum levels correlated with T-score of BMD.Conclusions Osteopenia and osteoporosis were common in SBS patients. There was a correlation between BMD and the serum levels of vitamins C, E and K, an indicative that such vitamins may influence bone health. Arch Endocrinol Metab. 2015;59(3):252-8.

OBJECTIVE: the present study was conducted to investigate the oxidant-antioxidant status in Egyptian children with sickle cell anemia. METHODS: the serum levels of total antioxidant capacity (TAO), paraoxonase (PON), vitamin E, nitrite, and malondialdehyde (MDA) were measured in 40 steady state children with homozygous sickle cell anemia (24 males and 16 females) and 20 apparently healthy age- and gender-matched controls. RESULTS: mean serum TAO, PON, vitamin E, and nitrite levels were significantly lower in the group with sickle cell anemia, whereas mean serum MDA was significantly higher in these children compared to controls. No significant differences in mean levels of TAO, PON, nitrite, vitamin E, and MDA were found in sickle cell anemia patients receiving hydroxyurea when compared with those not receiving hydroxyurea. A significant negative correlation between serum nitrite and the occurrence of vaso-occlusive crises (VOC) was observed (r = -0.3, p = 0.04). PON level was found to be positively correlated with patients' weight and BMI (r = -0.4, p = 0.01; r = -0.7, p < 0.001, respectively), but not with frequency of VOC. The area under the curve of serum nitrite in predicting occurrence of VOC was 0.782, versus 0.701 for PON, and 0.650 for TAO (p = 0.006). Serum MDA was not correlated with nitrite, PON, TAO, or vitamin E levels. No significant correlations were detected between serum nitrite and hemoglobin or antioxidant enzymes. CONCLUSION: children with sickle cell anemia have chronic oxidative stress that may result in increased VOC, and decreased serum nitrite may be associated with increases in VOC frequency. A novel finding in this study is the decrease in PON level in these patients, which is an interesting subject for further research. .


OBJETIVO: o presente estudo foi realizado com o objetivo de investigar o estado oxidante-antioxidante em crianc¸as egípcias com anemia falciforme. MÉTODOS: dosamos os níveis séricos da capacidade antioxidante total (CAT), paraoxonase (PON), vitamina E, nitrito e malondialdeído (MDA) em 40 crianças estáveis com anemia falciforme homozigótica (24 meninos e 16 meninas), e 20 controles pareados por idade/sexo aparente-mente saudáveis. RESULTADOS: os níveis séricos médios da CAT, PON, vitamina E e nitrito foram significativamente menores, ao passo que o nível sérico médio de MDA foi significativamente maior em crianças com anemia falciforme (AF), em comparação aos controles. Não foram encontradasdiferenças significativas nos níveis médios de CAT, PON, nitrito, vitamina E e MDA em pacientescom AF em tratamento com hidroxiureia, em comparação aos que receberam hidroxiureia. Encontramos uma correlação negativa significativa entre o nitrito sérico e a ocorrência decrises vaso-oclusivas agudas (CVO) (r = -0,3, p = 0,04). Descobrimos que o nível de PON está correlacionado positivamente com o peso e o IMC dos pacientes (r = -0,4; p = 0,01; r = -0,7; p < 0,001, respectivamente), porém não com a frequência de CVO. A área sob a curva (ASC) donitrito sérico na previsão da ocorrência de CVO foi 0,782, em comparação a 0,701 para PON e 0,650 para CAT (p = 0,006). O MDA não está correlacionado a nitrito, PON, CAT ou vitamina E. Não foram detectadas correlações significativas entre nitrito sérico e hemoglobina ou enzimas antioxidantes. OBJECTIVES: The aim of this study was to investigate neuropsychological performance and biomarkers of oxidative stress in patients with obstructive sleep apnea and the relationships between these factors. METHODS: This was an observational, cross-sectional study of 14 patients (36.0±6.5 years old) with obstructive sleep apnea and 13 controls (37.3±6.9 years old). All of the participants were clinically evaluated and underwent full-night polysomnography as well as neuropsychological tests. Blood samples were used to assay superoxide dismutase, catalase, glutathione and homocysteine, as well as vitamins E, C, B11 and B12. RESULTS: The patients performed poorly relative to the controls on several neuropsychological tests, such as the attention test and tests of long-term memory and working memory/executive function. They also had lower levels of vitamin E (p<0.006), superoxide dismutase (p<0.001) and vitamin B11 (p<0.001), as well as higher concentrations of homocysteine (p<0.02). Serum concentrations of vitamin C, catalase, glutathione and vitamin B12 were unaltered. Vitamin E levels were related to performance in the backward digit span task (F = 15.9; p = 0.002) and this correlation remained after controlling for age and body mass index (F = 6.3, p = 0.01). A relationship between superoxide dismutase concentrations and executive non-perseveration errors in the Wisconsin Card Sorting Test (F = 7.9; p = 0.01) was also observed. CONCLUSIONS: Decreased levels of antioxidants and lower performance on the neuropsychological tasks were observed in patients with obstructive sleep apnea. This study suggests that an imbalance between antioxidants and pro-oxidants may contribute to neuropsychological alterations in this patient population. .

Organophosphate (OP) pesticides such as dichlorvos (DDVP) intoxication has been shown to produce oxidative stress due to the generation of free radicals, which alter the antioxidant defense system in erythrocytes. In this study, the effects of DDVP (1, 10, 100 µM) or DDVP + vitamin C (VC; 10 µM) or vitamin E (VE; 30 µM), on the levels of malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activities in human erythrocytes were examined in vitro. There were no statistical differences between all groups for 1 µM concentration of DDVP. Treatment with DDVP alone produced an increase in the level of MDA and decreased activities of antioxidant enzymes (P < 0.05). Groups treated with vitamins and DDVP showed protective effects of vitamins against DDVP-induced changes in antioxidant enzyme activity and lipid peroxidation (LPO) (10 µM). At 100 µM concentration of DDVP vitamins had no effect on DDVP-induced toxicity. The results show that administration of DDVP resulted in the induction of erythrocyte LPO and alterations in antioxidant enzyme activities, suggesting that reactive oxygen species (ROS) may be involved in the toxic effects of DDVP. Also the data show that the plasma level of VC and VE may ameliorate OP-induced oxidative stress by decreasing LPO in erythrocytes at certain doses of OP pesicides.

PURPOSE: To evaluate whether alterations in plasma vitamin A and E levels in patients with psoriasis have an effect on tear film changes. METHODS: Sixty-two eyes of 31 patients with psoriasis vulgaris (Group A) and 74 eyes of 37 age- and gender-matched control subjects (Group B) were included in the study. Ocular and medical histories and dietary habits were obtained from each patient. The tear film break-up time (TBUT), the Schirmer 1 test results and plasma vitamin A and E levels were evaluated. RESULTS: The mean Schirmer 1 test score was 14.76 +/- 6.12 mm/5 min in Group A and 15.69 +/- 3.10 mm/5 min in Group B. The mean plasma levels of vitamins A and E in Groups A and B were 1.86 +/- 0.62 micromol/L and 1.88 +/- 0.65 micromol/L vs. 26.21 +/- 5.13 micromol/L and 27.19 +/- 8.89 micromol/L, respectively. The Schirmer 1 test results and plasma vitamin A and E levels were not found to be significantly different between the groups (p > 0.05). The mean TBUT was 9.94 +/- 6.18 seconds in Group A and 14.47 +/- 5.65 seconds in Group B, a significant difference (p 0.05). CONCLUSIONS: Plasma vitamin A and E levels do not seem to be related to tear film changes in patients with psoriasis vulgaris.

OBJETIVOS: Diagnosticar bioquimicamente o estado nutricional de vitamina E de lactantes por meio da análise do alfa-tocoferol no soro e no colostro, verificar sua associação com variáveis maternas e determinar a prevalência de deficiência de vitamina E nessas mulheres. MÉTODOS: Participaram do estudo 103 puérperas que foram classificadas quanto às seguintes variáveis maternas: idade, estado nutricional pré-gestacional, ganho de peso gestacional, paridade e tipo de parto. Amostras de soro e colostro foram coletadas em jejum no pós-parto imediato e o alfa-tocoferol foi analisado por cromatografia líquida de alta eficiência (CLAE). Para definir o estado nutricional de vitamina E, foi adotado ponto de corte sérico (697,7 μg/dL). A análise estatística foi realizada com o uso do teste t de Student para amostras independentes e correlação de Pearson. As diferenças foram consideradas significativas quando p<0,05. RESULTADOS: A concentração média de alfa-tocoferol foi 1.125,1±551,0 μg/dL no colostro e 1.138,6±346,0 μg/dL no soro, indicativo de estado nutricional bioquímico adequado. Entretanto, ao analisar as puérperas individualmente, constatamos que 16% apresentaram valores abaixo do esperado para esta vitamina. As mulheres submetidas à cesárea apresentaram níveis de alfa-tocoferol no colostro (1.280±591 μg/dL) significativamente maiores em relação àquelas cuja via de parto foi a normal (961,7±370 μg/dL) (p<0,05). Verificou-se que as parturientes com excesso de peso pré-gestacional tiveram concentração da vitamina no colostro maior (1.331,5±548 μg/dL) quando comparadas às mulheres com baixo peso (982,1±374 μg/dL) ou eutrofia (992,3±346 μg/dL) (p<0,05). Entretanto, as demais variáveis estudadas não apresentaram associação com o alfa-tocoferol do colostro. Além disso, nenhuma variável mostrou estar relacionada aos níveis da vitamina no soro materno e não foi demonstrada correlação entre os níveis de alfa-tocoferol no soro e no leite. CONCLUSÕES: Apesar do diagnóstico de satisfatório estado nutricional, as lactantes apresentaram risco importante de deficiência subclínica para vitamina E. Sugere-se que a concentração de alfa-tocoferol presente no colostro esteja associada ao tipo de parto e ao estado nutricional pré-gestacional da mulher.


PURPOSE: To determine the nutritional status of vitamin E in breastfeeding women through the analysis of alpha-tocopherol concentration in serum and colostrum, to analyze its relation with maternal variables and to determine the prevalence of vitamin E deficiency in these women. METHODS: The study included 103 mothers who were classified according to maternal variables: age, nutritional status before pregnancy, gestational weight gain, parity and mode of delivery. Colostrum and serum samples were collected under fasting conditions in the immediate postpartum period. Alpha-tocopherol was analyzed by high performance liquid chromatography (HPLC). A serum cutoff of 697.7 μg/dL was adopted to define the nutritional status of vitamin E. Statistical analysis was performed with the Student's t test for independent samples and Pearson's correlation. Differences were significant when p<0.05. RESULTS: The average concentration of alpha-tocopherol was 1.125±551.0 μg/dL in colostrum and 1,138.6±346.0 μg/dL in serum, indicating adequate biochemical nutritional status. However, when analyzing the mothers individually, a 16% rate of subclinical vitamin E deficiency was detected. Women undergoing cesarean delivery had significantly higher alpha-tocopherol levels in colostrum (1.280±591 μg/dL) compared with those undergoing normal delivery (961.7±370 μg/dL) (p<0.05). It was found that mothers who were overweight before pregnancy had higher vitamin concentration in colostrum (1,331.5±548 μg/dL) when compared to underweight women (982.1±374 μg/dL) or women of normal weight (992.3±346 μg/dL) (p<0.05). However, the other variables were not associated with alpha-tocopherol in colostrum. Moreover, no variable showed association with vitamin E levels in maternal serum and no correlation was demonstrated between the alpha-tocopherol levels in serum and in milk. CONCLUSIONS: Despite the diagnosis of satisfactory nutritional status, breastfeeding women showed significant risk of subclinical vitamin E deficiency. We suggest that the concentration of alpha-tocopherol in colostrum be associated with type of delivery and pre-gestational nutritional status of women.

ABSTRACT

Background: Homozygous β thalassemia may lead to a marked reduction or absence of normal β chain production and accumulation of unpaired alpha-globin chains. A crucial component in the oxidant susceptibility of the thalassemic RBC is the release of heme and iron from the excessive, unpaired α-globin chains. This release can initiate self-amplifying redox reactions, which deplete the cellular reduction potential (e.g., GSH), oxidize additional hemoglobin and accelerate RBC destruction. Furthermore, β-thalassemia patients are under continuous blood transfusion, which, although life-saving, leads to an iron overload with a resultant increase in non-transferrin-bound iron that may cause greater tissue toxicity than iron in other forms. Iron-induced oxidative stress is known to be one of the most important factors determining cell injury in thalassemic patients. Therefore, we designed this study to obtain a comprehensive picture of the iron overload, antioxidant status and cell damage in β thalassemia major patients undergoing regular blood transfusion. Materials and Methods: A total of 48 diagnosed patients of β thalassemia major and 30 age- and sex-matched healthy subjects were included in the study. Estimation of hemoglobin, hematocrit, glutathione peroxidase (GPX), superoxide dismutase (SOD),vitamin E, serum ferritin, total and direct bilirubin, AST and ALT was carried out. Results: The levels of vitamin E, antioxidant enzymes GPX and SOD were significantly lowered in β thalassemic patients as compared with the control group (P<0.001). Serum total and direct bilirubin, AST and ALT were significantly elevated in thalassemic subjects as compared with the control group, indicating liver cell damage. Conclusion: Thus, our findings indicate that thalassemics are in a state of enhanced oxidative stress and that the administration of selective antioxidants would represent a promising approach toward counteracting oxidative damage and its deleterious effects on the disease status.

Background & objectives: The human system possesses antioxidants that act harmoniously to neutralize the harmful oxidants. This study was aimed to evaluate the serum total antioxidant capacity (TAC) as a single parameter in Eales’ disease (ED) and in an acute inflammatory condition such as uveitis and in cataract which is chronic, compared to healthy controls. Methods: The TAC assay was done spectrophotometrically in the serum of Eales’ disease cases (n=20) as well as in other ocular pathologies involving oxidative stress namely, uveitis and cataract (n=20 each). The oxidative stress measured in terms of TBARS, was correlated with the TAC. Individual antioxidants namely vitamin C, E and glutathione were also estimated and correlated with TAC. Results: TAC was found to be significantly lower in Eales’ disease with active vasculitis (0.28 ± 0.09 mM, P<0.001), Eales’ disease with healed vasculitis (0.67 ± 0.09 mM), uveitis (0.46 ± 0.09 mM, P<0.001) and cataract (0.53 ± 0.1 mM, P=0.001) compared to the healthy controls, with a TAC level of 0.77 ± 0.09 mM. The TAC was found to correlate positively with vitamin E levels (P=0.05), GSH (P=0.02) but not with vitamin C, as seen in ED cases. In ED cases supplemented with vitamin E and C, there was a significant increase in the TAC level (P=0.02). Interpretation & conclusions: The TAC measurement provided a comprehensive assay for establishing a link between the antioxidant capacity and the risk of disease as well as monitoring antioxidant therapy. This method is a good substitute for assay of individual antioxidants as it clearly gives the status of the oxidative stress in the disease process.

ABSTRACT

Many studies have investigated the possible role of reactive oxygen species in the etiology and pathogenesis of Rheumatoid Arthritis [RA]. The aim of this study was to investigate the activities of some antioxidants in RA patients. In this case-control study, 59 RA patients and 60 healthy sex and age-matched controls were selected. Vitamin E and Beta-carotene were determined using HPLC. Erythrocytes glutathione reductase [GR] activity was measured spectrophotometrically, and malondialdehyde [MDA] was determined by colorimetric method. Arylesterase activity [AEA] was measured by Phenylacetate. The clinical data were determined by a rheumatologist, medical history and filling the questionnaire by interview. Statistical analyses were carried out using the SPSS software. In patients with RA, serum MDA level was significantly higher and plasma concentration of vitamin E, Beta-carotene and GR activity, were significantly lower than healthy control [P<0.001]. AEA activity differences between two groups were non-significant. Oxidative stress may play an important role in the inflammation and pathogenesis of RA

ABSTRACT

Giardia lamblia, a common parasite of the human small bowel, is responsible for diarrhea, nutritional deficiencies and production of free radicals. Micronutrients regulate many physiological functions and participate in antioxidant defense. Changes in their serum levels occur in several gastrointestinal disorders. The present study aimed to investigate the homeostasis of some micronutrients and antioxidant defense in human giardiasis. The study was designed to compare serum levels of some micronutrients and antioxidant status in 30 patients with symptomatic giardiasis and 30 age- and sex-matched controls. Giardia trophozoites and/or cysts were detected in the stool samples of patients using saline and iodine wet mount preparations and formol ether technique. Levels of magnesium, iron, manganese, copper and chromium were measured using atomic absorption spectrophotometer. Vitamin E level was determined as well as total antioxidant capacity [TAC] and serum levels of the antioxidant biomarkers; albumin, total bilirubin and uric acid were evaluated. Giardia-infected patients displayed significantly lower levels of magnesium, iron, manganese, chromium and vitamin E and higher levels of copper and uric acid compared to controls. TAC, albumin and bilirubin in Giardia-infected patients were not significantly different from the controls. The pathophysiological changes in giardiasis alter the homeostasis of some micronutrients. Uric acid constitutes an important antioxidant defense in human giardiasis that may compensate for the deficiency of some antioxidant micronutrients. Prevention and treatment of giardiasis may have a positive impact on the micronutrient status and on the general health of the population

As deficiências de vitaminas, verificadas em indivíduos com HIV/Aids em terapia antirretroviral de alta potência (HAART) têm sido associadas à piora do curso clínico da doença e maior risco de mortalidade. Objetivo Mostrar a distribuição das concentrações séricas de vitaminas A e E, e beta-caroteno em adultos com infecção pelo HIV/Aids e estudar a associação de suas concentrações, segundo diferentes esquemas de HAART. Métodos Foram selecionados 182 adultos de 20 a 59 anos de idade, de ambos os sexos, com HIV/Aids em HAART estável por no mínimo 6 meses, e com níveis de linfócitos T-CD4+ 200 células/mm3. Os indivíduos foram divididos em três grupos por esquema de HAART utilizado: inibidores de transcriptase reversa análogos de nucleosídeo (ITRN) associados a inibidores de transcriptase reversa não análogos de nucleosídeo (ITRNN); ITRN associada a inibidores de protease (IP); ITRN associadas a outras classes (inibidores de fusão, inibidores de integrasse, inibidores de entrada e IP associada a essas medicações). A determinação dos micronutrientes foi realizada por cromatografia líquida de alta eficiência. Foram verificadas variáveis sócio-demográficas e econômicas, estilo de vida, história da doença, uso de medicações e variáveis antropométricas e laboratoriais. Para medir os efeitos das variáveis explanatórias (esquemas de tratamento, tempo de uso e adesão ao último esquema) sobre cada variável resposta (retinol, alfa-tocoferol e beta-caroteno), foram realizadas análises de regressão linear múltipla. Sexo, idade, escolaridade, tabagismo, prática de atividade física, tempo de infecção por HIV, presença de comorbidades, relação cintura-quadril e níveis de linfócitos T-CD4+ e colesterol foram usadas como variáveis de controle. Deficiency of vitamins found in individuals with HIV/AIDS on highly active antiretroviral therapy (HAART) has been associated with an increased risk of disease progression and mortality. Objective To show the distribution of serum concentrations of vitamins A and E and beta-carotene in adults with HIV/AIDS, and to study the association of their concentrations, according to different regimens of HAART. Methods We selected 182 men and women aged 20-59 years with HIV/AIDS on stable HAART for at least six months and with levels of CD4+ T-lymphocytes 200 cells/mm3. Individuals were divided into three groups according to the HAART regimen used: nucleoside reverse transcriptase inhibitors (NRTI) combined with nonnucleoside reverse transcriptase inhibitors (NNRTI); NRTI combined with protease inhibitors (PI); NRTI combined with other classes (fusion inhibitors, integrase inhibitors, entry inhibitors, and PI associated with these medications). Determinations of vitamins A and E and beta-carotene were performed by high-performance liquid chromatography. Socio-demographic and economic variables, lifestyle, disease history, medication use, and anthropometric and laboratory variables were assessed. Multiple regression analyses were used to measure the effects of the explanatory variables (treatment regimens, duration and adherence to the last treatment regimen) on each response variable (retinol, alpha-tocopherol and beta-carotene). Polycystic ovary syndrome Polycystic Ovarian Syndrome is a disease characterized by unovulation and formation of immature follicles result in menstrual irregularities, hyperandrogenemia, hirsutism and infertility. This study was aimed to show the changes in oxidative status in these women, in accordance to changes in serum antioxidant vitamins A,C,E. Thirty Polycystic Ovarian Syndrome women included in this study, their serum FSH, LH, Prolactin and testosterone hormones were measured .also serum concentrations of vitamin A, C, E were determined. These women are divided into three groups according to their hormonal changes. Group [1] involves women with high level of prolactin. Group [2] involves women with LH/FSH ratio more than one. Group [3] involves women with LH/FSH ratio less than one. All groups are compared with 10 apparently healthy fertile women. All groups show significant decrease in serum concentrations of vitamins in compare with control women .This indicates different hormonal changes increase oxidative stress in Polycystic Ovarian Syndrome women by different mechanisms. To examine the serum antioxidant levels like vit-C, vit-E and glutathione in patients with renal diseases who were subjected to dialysis and to evaluate the antioxidant by FRAP method. To find out the involvement of free radicals in pathogenesis of renal disease. Fifty patients with high levels of creatinine and urea level were included in the study of dialysis. A difference of antioxidant level of vit-C, vit-E and glutathione was observed. The study therefore suggests the importance of assessing these marker oxidative stress antioxidant capacities in renal disease during dialysis

ABSTRACT

To investigate biochemical changes in lipid peroxidation, nitric oxide, and vitamin E in recurrent pregnancy loss women, and compare these with healthy pregnant, and non-pregnant women. A case control study was conducted from September 2008 to December 2009 at Al Khadimiya Teaching Hospital, Baghdad, Iraq. Ninety-six subjects were included in the study, 32 were patients with recurrent pregnancy loss [RPL], and 32 pregnant women in their third trimester, and another 32 non-pregnant women were used as controls. Blood samples were collected from each patient at the time of pregnancy loss. Serum from patients and controls was then used to estimate malondialdehyde [MDA], nitric oxide [NO], and vitamin E levels. There was a significant elevation in patient serum MDA compared with third trimester pregnant women [p=0.002] and non-pregnant women [p=0.0001]. Both serum vitamin E and NO levels in RPL patients also showed a highly significant decrease compared with third trimester pregnant, and non-pregnant women. A highly significant difference was found in the MDA/vitamin E ratio between RPL and control groups, while no significance was found between RPL and control groups' NO/vitamin E ratio. The decrease in NO production and vitamin E is a result of RPL and not a causative factor, as the RPL was without pathological cause, medication, or fibroid presence, and no significant difference was found between the NO/vitamin E ratio in RPL and controls group

Background: Free radical Injury is associated with cancer, but how the extent of oxidative stress correlates with the FIGO (International Federation of Gynecology and Obstetrics) stage in Carcinoma Cervix (Ca Cx), and its significance as a prognostic marker, is not clear and needs an in-depth study. Aim: To correlate the blood levels of Lipid Peroxidation (LPO), Reduced Glutathione (GSH), Superoxide Dismutase (SOD), and Vitamin A and E levels with the clinical stage in Ca Cx. Settings and Design: This is a Prospective Case Control Study. Materials and Methods: LPO, SOD, reduced GSH were estimated by Bio Chemical Assays and Vitamins by High Performance Liquid Chromatography (HPLC). Statistical Analysis: The cases and controls were compared using One Way ANOVA and different stages over different time periods were individually compared by Repeated Measure Analysis of Variance. Results: The results indicated a statistically significant increase of LPO vis-a-vis the FIGO stage of Ca Cx and control, while the antioxidant status as depicted by GSH and SOD decreased. Vitamin A and E levels were significantly lower in cancer cases as compared to the control. Conclusion: Increased LPO and reduced antioxidant levels may be taken as associated predictive markers, thus suggesting that Ca Cx cases should get nutritive supplements to contain the blood LPO level and maintain a positive balance of antioxidants for a better outcome in terms of delayed recurrence and better Quality of Life (QOL).

La Fenilquetonuria (PKU) se produces por la deficiencia de la enzima Fenilalanina Hidroxilasa, causando un aumento plasmático de fenilalanina (FA). El objetivo del presente estudio fue evaluar la ingesta de Selenio (Se), Zinc (Zn) y vitamina E en niños PKU menores de 13 años de edad, que estaban en dieta restringida en FA y recibiendo fórmula especial sin FA. Método: Se incluyeron 50 PKU entre los 0 y 13 años y en control en el INTA, Universidad de Chile. Se analizó ingesta de vitamina E, Se y Zn, se midió nivel de FA en plasma y se evaluó estado nutritional. Resultados: Las recomendaciones diarias de Se y Zn se cubren en un 100 por ciento con la dieta habitual de PKU. Pero al excluir la formula sin FA, la cobertura de ambos nutriente disminuye a 45 por ciento y 20 por ciento respectivamente. La ingesta de vitamina E se cubre en forma natural por el consumo de aceites vegetales y al incluir la vitamina de la fórmula sin FA, la cobertura se incrementaba 5 veces sobre su recomendación. El nivel de FA en la sangre fue en promedio de 5.4 mg/dL, considerado un buen control metabólico. El 64 por ciento tenía un estado nutritional normal, el 30 por ciento estaba sobrepeso u obeso y un 6 por ciento riesgo de desnutrición. Conclusiones: Se concluye que la dieta de niños PKU cubre las recomendaciones de los micronutrientes: Zn, Se y vitamina E. No obstante se debe enfatizar la importancia que tiene la fórmula sin FA para cumplir con las recomendaciones nutricionales, especialmente de micronutrientes.


Phenylketonuria (PKU) is caused by a deficiency of the enzyme Phenylalanine Hydroxylase, resulting in increased plasma Phenylalanine (Phe). The aim of this study was to assess the intake of Selenium (Se), Zinc (Zn) and vitamin E in PKU children, who were on a diet restricted in Phenylalanine (Phe) and receiving a special formula without Phe. Method: the study included 50 PKU children between 0 and 13 years controled at INTA, University of Chile. We analyzed intake of vitamin E, Se and Zn, measured Phe plasma levels and assessed nutritional status. Results: The daily recommendations of Se and Zn were 100 percent covered with the usual PKU diet. By excluding the formula without Phe, the coverage of both nutrients decreased to 45 percent and 20 percent respectively. The intake of vitamin E was covered by the consumption of vegetable oils and when the formula without Phe was included, the coverage was increased 5 times over the recommended levels. Blood Phe level remained on average at 5.4 mgldL, considered a good metabolic control. Conclusions: We conclude that the diet of PKU children covered the recommended levels of Zn, Se and vitamin E. However, it should be emphasized the importance of the formula without Phe to meet nutritional recommendations, particularly of micronutrients.
